Description
Isolated actinomycetes using the serial dilution plate approach. To test soil samples 1 g was suspended in 10 ml of sterile normal saline (0.85%) and agitated on an orbital shaker for 30 minutes at 121 rpm. The suspension was then put in the Laminar Air Flow hood for 30 minutes. To dilute the suspension 7-fold, 1 ml was taken and added to 9 ml of sterile normal saline (0.85%). After vortexing, 0.1ml of suspension from 10-3, 10-5, and 10-7 dilutions was dispersed on the Starch Casein Agar medium and Actinomycetes Isolation Agar medium. The plates were incubated at 30°C for 7-10 days
